Conventional hematoxylin-eosin (H&E) staining is limited to revealing cell morphology and distribution, whereas immunohistochemical (IHC) staining provides precise and specific visualization of protein activation at the molecular level. Virtual staining technology has emerged as a solution for highly efficient IHC examination, which directly transforms H&E-stained images to IHC-stained images. However, virtual staining is challenged by the insufficient mining of pathological semantics and the spatial misalignment of pathological semantics. To address these issues, we propose the Pathological Semantics-Preserving Learning method for Virtual Staining (PSPStain), which directly incorporates the molecular-level semantic information and enhances semantics interaction despite any spatial inconsistency. Specifically, PSPStain comprises two novel learning strategies: 1) Protein-Aware Learning Strategy (PALS) with Focal Optical Density (FOD) map maintains the coherence of protein expression level, which represents molecular-level semantic information; 2) Prototype-Consistent Learning Strategy (PCLS), which enhances cross-image semantic interaction by prototypical consistency learning. We evaluate PSPStain on two public datasets using five metrics: three clinically relevant metrics and two for image quality. Extensive experiments indicate that PSPStain outperforms current state-of-the-art H&E-to-IHC virtual staining methods and demonstrates a high pathological correlation between the staging of real and virtual stains.

Digital Subtraction Angiography (DSA) is one of the gold standards in vascular disease diagnosing. With the help of contrast agent, time-resolved 2D DSA images deliver comprehensive insights into blood flow information and can be utilized to reconstruct 3D vessel structures. Current commercial DSA systems typically demand hundreds of scanning views to perform reconstruction, resulting in substantial radiation exposure. However, sparse-view DSA reconstruction, aimed at reducing radiation dosage, is still underexplored in the research community. The dynamic blood flow and insufficient input of sparse-view DSA images present significant challenges to the 3D vessel reconstruction task. In this study, we propose to use a time-agnostic vessel probability field to solve this problem effectively. Our approach, termed as vessel probability guided attenuation learning, represents the DSA imaging as a complementary weighted combination of static and dynamic attenuation fields, with the weights derived from the vessel probability field. Functioning as a dynamic mask, vessel probability provides proper gradients for both static and dynamic fields adaptive to different scene types. This mechanism facilitates a self-supervised decomposition between static backgrounds and dynamic contrast agent flow, and significantly improves the reconstruction quality. Our model is trained by minimizing the disparity between synthesized projections and real captured DSA images. We further employ two training strategies to improve our reconstruction quality: (1) coarse-to-fine progressive training to achieve better geometry and (2) temporal perturbed rendering loss to enforce temporal consistency. Experimental results have demonstrated superior quality on both 3D vessel reconstruction and 2D view synthesis.

Chest X-rays (CXRs) are commonly utilized as a low-dose modality for lung screening. Nonetheless, the efficacy of CXRs is somewhat impeded, given that approximately 75% of the lung area overlaps with bone, which in turn hampers the detection and diagnosis of diseases. As a remedial measure, bone suppression techniques have been introduced. The current dual-energy subtraction imaging technique in the clinic requires costly equipment and subjects being exposed to high radiation. To circumvent these issues, deep learning-based image generation algorithms have been proposed. However, existing methods fall short in terms of producing high-quality images and capturing texture details, particularly with pulmonary vessels. To address these issues, this paper proposes a new bone suppression framework, termed BS-Diff, that comprises a conditional diffusion model equipped with a U-Net architecture and a simple enhancement module to incorporate an autoencoder. Our proposed network cannot only generate soft tissue images with a high bone suppression rate but also possesses the capability to capture fine image details. Additionally, we compiled the largest dataset since 2010, including data from 120 patients with high-definition, high-resolution paired CXRs and soft tissue images collected by our affiliated hospital. Extensive experiments, comparative analyses, ablation studies, and clinical evaluations indicate that the proposed BS-Diff outperforms several bone-suppression models across multiple metrics. Our code can be accessed at https://github.com/Benny0323/BS-Diff.

Deep learning in digital pathology brings intelligence and automation as substantial enhancements to pathological analysis, the gold standard of clinical diagnosis. However, multiple steps from tissue preparation to slide imaging introduce various image corruptions, making it difficult for deep neural network (DNN) models to achieve stable diagnostic results for clinical use. In order to assess and further enhance the robustness of the models, we analyze the physical causes of the full-stack corruptions throughout the pathological life-cycle and propose an Omni-Corruption Emulation (OmniCE) method to reproduce 21 types of corruptions quantified with 5-level severity. We then construct three OmniCE-corrupted benchmark datasets at both patch level and slide level and assess the robustness of popular DNNs in classification and segmentation tasks. Further, we explore to use the OmniCE-corrupted datasets as augmentation data for training and experiments to verify that the generalization ability of the models has been significantly enhanced.

Fundus image captures rear of an eye, and which has been studied for the diseases identification, classification, segmentation, generation, and biological traits association using handcrafted, conventional, and deep learning methods. In biological traits estimation, most of the studies have been carried out for the age prediction and gender classification with convincing results. However, the current study utilizes the cutting-edge deep learning (DL) algorithms to estimate biological traits in terms of age and gender together with associating traits to retinal visuals. For the traits association, our study embeds aging as the label information into the proposed DL model to learn knowledge about the effected regions with aging. Our proposed DL models, named FAG-Net and FGC-Net, correspondingly estimate biological traits (age and gender) and generates fundus images. FAG-Net can generate multiple variants of an input fundus image given a list of ages as conditions. Our study analyzes fundus images and their corresponding association with biological traits, and predicts of possible spreading of ocular disease on fundus images given age as condition to the generative model. Our proposed models outperform the randomly selected state of-the-art DL models.

Pathologists need to combine information from differently stained pathology slices for accurate diagnosis. Deformable image registration is a necessary technique for fusing multi-modal pathology slices. This paper proposes a hybrid deep feature-based deformable image registration framework for stained pathology samples. We first extract dense feature points via the detector-based and detector-free deep learning feature networks and perform points matching. Then, to further reduce false matches, an outlier detection method combining the isolation forest statistical model and the local affine correction model is proposed. Finally, the interpolation method generates the deformable vector field for pathology image registration based on the above matching points. We evaluate our method on the dataset of the Non-rigid Histology Image Registration (ANHIR) challenge, which is co-organized with the IEEE ISBI 2019 conference. Our technique outperforms the traditional approaches by 17% with the Average-Average registration target error (rTRE) reaching 0.0034. The proposed method achieved state-of-the-art performance and ranked 1st in evaluating the test dataset. The proposed hybrid deep feature-based registration method can potentially become a reliable method for pathology image registration.

This paper revisits the fundamental mathematics of Taylor series to approximate curves with function representation and arc-length-based parametric representation. Parametric representation is shown to preserve its form in coordinate transformation and parameter shifting. These preservations can significantly facilitate lane estimation in vehicle control since lanes perceived by cameras are typically represented in vehicle body-fixed frames which are translating and rotating. Then we derived the transformation from function representation to arc-length-based parametric representation and its inverse. We applied the transformation to lane estimation in vehicle control problem, and derived the evolution of coefficients for parametric representation that can be used for prediction. We come up with a procedure to simulate the whole process with perception, lane estimation and control for the path-following problem. Simulations are performed to demonstrate the efficacy of the proposed lane estimation algorithm using parametric representation. The results indicate that the proposed technique ensures that vehicle control can achieve reasonably good performance at very low perception updating rate.

This paper investigates the lateral control problem in vehicular path-following when the feedback sensor(s) are mounted at an arbitrary location in the longitudinal symmetric axis. We point out that some existing literature has abused the kinematic bicycle model describing the motion of rear axle center for other locations, which may lead to poor performance in practical implementations. A new nonlinear controller with low-complexity and high-maneuverability is then designed that takes into account senor mounting location, driving comfort and transient response with large initial errors. Design insights and intuitions are also provided in detail. Furthermore, analysis on stability and tracking performance for the closed-loop system are studied, and conditions and guidelines are provided on the selection of control parameters. Comprehensive simulations are performed to demonstrate the efficacy of the proposed nonlinear controller for arbitrary sensor locations. Meanwhile, we also show that designing controllers ignoring the sensor location may lead to unexpected vehicular sway motion in non-straight paths.

Reducing the radiation exposure for patients in Total-body CT scans has attracted extensive attention in the medical imaging community. Given the fact that low radiation dose may result in increased noise and artifacts, which greatly affected the clinical diagnosis. To obtain high-quality Total-body Low-dose CT (LDCT) images, previous deep-learning-based research work has introduced various network architectures. However, most of these methods only adopt Normal-dose CT (NDCT) images as ground truths to guide the training of the denoising network. Such simple restriction leads the model to less effectiveness and makes the reconstructed images suffer from over-smoothing effects. In this paper, we propose a novel intra-task knowledge transfer method that leverages the distilled knowledge from NDCT images to assist the training process on LDCT images. The derived architecture is referred to as the Teacher-Student Consistency Network (TSC-Net), which consists of the teacher network and the student network with identical architecture. Through the supervision between intermediate features, the student network is encouraged to imitate the teacher network and gain abundant texture details. Moreover, to further exploit the information contained in CT scans, a contrastive regularization mechanism (CRM) built upon contrastive learning is introduced.CRM performs to pull the restored CT images closer to the NDCT samples and push far away from the LDCT samples in the latent space. In addition, based on the attention and deformable convolution mechanism, we design a Dynamic Enhancement Module (DEM) to improve the network transformation capability.

Feature selection is important in data representation and intelligent diagnosis. Elastic net is one of the most widely used feature selectors. However, the features selected are dependant on the training data, and their weights dedicated for regularized regression are irrelevant to their importance if used for feature ranking, that degrades the model interpretability and extension. In this study, an intuitive idea is put at the end of multiple times of data splitting and elastic net based feature selection. It concerns the frequency of selected features and uses the frequency as an indicator of feature importance. After features are sorted according to their frequency, linear support vector machine performs the classification in an incremental manner. At last, a compact subset of discriminative features is selected by comparing the prediction performance. Experimental results on breast cancer data sets (BCDR-F03, WDBC, GSE 10810, and GSE 15852) suggest that the proposed framework achieves competitive or superior performance to elastic net and with consistent selection of fewer features. How to further enhance its consistency on high-dimension small-sample-size data sets should be paid more attention in our future work. The proposed framework is accessible online (https://github.com/NicoYuCN/elasticnetFR).

Computed Tomography (CT) is an advanced imaging technology used in many important applications. Here we present a deep-learning (DL) based CT super-resolution (SR) method that can reconstruct low-resolution (LR) sinograms into high resolution (HR) CT images. The method synergistically combines a SR model in sinogram domain, a deblur model in image domain, and the iterative framework into a CT SR algorithm super resolution and deblur based iterative reconstruction (SADIR). We incorporated the CT domain knowledge into the SADIR and unrolled it into a DL network (SADIR Net). The SADIR Net is a zero shot learning (ZSL) network, which can be trained and tested with a single sinogram in the test time. The SADIR was evaluated via SR CT imaging of a Catphan700 physical phantom and a biological ham, and its performance was compared to the other state of the art (SotA) DL-based methods. The results show that the zero-shot SADIR-Net can indeed provide a performance comparable to the other SotA methods for CT SR reconstruction, especially in situations where training data is limited. The SADIR method can find use in improving CT resolution beyond hardware limits or lowering requirement on CT hardware.

In this paper, we develop a mechanical system inspired microscopic traffic model to characterize the longitudinal interaction dynamics among a chain of vehicles. In particular, we extend our prior work on mass-spring-damper-clutch based car-following model between two vehicles to multi-vehicle scenario. This model can naturally capture the driver's tendency to maintain the same speed as the vehicle ahead while keeping a (speed-dependent) desired spacing. It is also capable of characterizing the impact of the following vehicle on the preceding vehicle, which is generally neglected in existing models. A new string stability criterion is defined for the considered multi-vehicle dynamics, and stability analysis is performed on the system parameters and time delays. An efficient online parameter identification algorithm, sequential recursive least squares with inverse QR decomposition (SRLS-IQR), is developed to estimate the driving-related model parameters. These real-time estimated parameters can be employed in advanced longitudinal control systems to enable accurate prediction of vehicle trajectories for improved safety and fuel efficiency. The proposed model and the parameter identification algorithm are validated on NGSIM, a naturalistic driving dataset, as well as our own connected vehicle driving data. Promising performance is demonstrated.
