Hopkinton village is located in southwestern Middlesex County, at the center of the town of Hopkinton. [[Massachusetts Route 135]] passes through the community as Main Street, leading northeastward {{convert|6|mi|0}} to [[Framingham, Massachusetts|Framingham]] and northwest the same distance to [[Westborough (CDP), Massachusetts|Westborough]]. [[Massachusetts Route 85]] runs through the village as Cedar Street and Grove Street, leading north {{convert|5|mi|0}} to [[Southborough, Massachusetts|Southborough]] and south {{convert|6|mi|0}} to [[Milford (CDP), Massachusetts|Milford]]. [[Interstate 495 (Massachusetts)|Interstate 495]] passes {{convert|1|mi}} west of the village, with access from Route 30. I-495 leads north {{convert|9|mi|0}} to [[Marlborough, Massachusetts|Marlborough]] and southeast {{convert|12|mi}} to [[Franklin, Massachusetts|Franklin]].
